This lab tested the ability of plant cells to carry out photosynthesis in varying intensities of light. One dish of leaf disks was placed in the dark, one dish under the room lights, and one dish 25 cm from a light source. To measure photosynthetic activity, leaf disks were cut from spinach leaves and the oxygen stored in the leaves was removed by applying a vacuum. This caused the leaves to sink to the bottom of the flask. Leaf disks that sunk to the bottom of the flask were transferred to petri dishes containing bicarbonate.

When exposed to adequate light, photosynthesis occurred in the leaf disks. The leaf disks used bicarbonate as a source of carbon dioxide for photosynthesis and produced oxygen as a bi-product. Oxygen production, and thus photosynthesis, was indicated by floating leaf disks.
